چکیده

Tongluoxingnao (TLXN) effervescent tablets consist of Scutellaria baicalensis Georgi, Radix Angelicae Sinensis, and Ligusticum chuanxiong Hort that can improve cognitive dysfunction in dementia. This study aimed to evaluate the effect of TLXN on learning and memory impairment in vascular dementia (VD) rats’ model, explore the effect of the MAPK signaling pathway on chronic ischemia, and determine the underlying mechanism. VD was induced in rats by permanent occlusion of the bilateral common carotid arteries. The Morris water maze and histopathological examinations were used to evaluate the effect of TLXN on learning and memory impairment, whereas real-time PCR and Western blot analysis were used to determine the levels of mRNA and phosphorylation of members of the MAPK signaling pathway, respectively. Compared to rats in the sham group, VD rats exhibited learning and memory impairments such as escape latency prolongation and decreases in the times of entrance into the first quadrant, valid region and escape platform; however, these conditions improved following TLXN administration. Moreover, TLXN may regulate the p38 MAPK and extracellular signal-regulated kinase (ERK) signaling pathways via the inhibition of the phosphorylation of p38 MAPK (p38), ERK1/2, MAP kinase kinase 6 (MKK6), and c-jun and a reduction in the mRNA levels of p38, ERK1/2, and c-jun. These results suggest that TLXN demonstrates the potential for development as a drug for vascular dementia and that the underlying mechanism involves the regulation of the p38 and ERK MAPK signaling pathways.

The effects of crocin on spatial memory impairment induced by hyoscine: Role of NMDA, AMPA, ERK, and CaMKII proteins in rat hippocampus

Objective(s): Crocus sativus L. and its active constituent, crocin, have neuroprotective effects. The effects of crocin on memory impairment have been mentioned in studies but the signaling pathways  have not been evaluated. Therefore, the aim of this study was to evaluate the effects of crocin on the hyoscine-induced memory impairment in rat. Additionally, the level o...

Cinnamaldehyde improves methamphetamine-induced spatial learning and memory deficits and restores ERK signaling in the rat prefrontal cortex

Objective(s): Methamphetamine is a stimulant compound that penetrates readily into the central nervous system. Repeated exposure to methamphetamine leads to damage in the dopaminergic and serotonergic axons of selected brain regions. Previous studies showed that cinnamaldehyde improved memory impairment in animals. In the present study, we aimed to elucidate the effects of cinnamaldehyde on met...

Tanshinone IIA inhibits AGEs-induced proliferation and migration of cultured vascular smooth muscle cells by suppressing ERK1/2 MAPK signaling

Objective(s): Vascular smooth muscle cells (VSMCs) play a key role in the pathogenesis of diabetic vascular disease. Our current study sought to explore the effects of tanshinone IIA on the proliferation and migration of VSMCs induced by advanced glycation end products (AGEs). Materials and Methods: In this study, we examined the effects of tanshinone IIA by cell proliferation assay and cell mi...

Inactivation of mitogen-activated protein kinase signaling pathway reduces caspase-14 expression in impaired keratinocytes

Objective(s):Several investigations have revealed that caspase-14 is responsible for the epidermal differentiation and cornification, as well as the regulation of moisturizing effect. However, the precise regulation mechanism is still not clear. This study was aimed to investigate the expression of caspase-14 in filaggrin-deficient normal human epidermal keratinocytes (NHEKs) and to explore the...

P-111: EGFR, ERK, MEK Genes Expression Level in Cumulus Cells of PCOS Women Compared with Healthy Women

Background Poly cystic ovarian syndrome (PCOS) is known as a common endocrine disorder in women at reproductive ages and may cause developmental abnormality in oocyte. ERK has found as a regulator protein of Gap junctions (GJ) function and the level of exchanges between two neighbors cells, for example oocyte and surrounding cumulus cells (CCs) in the mammalian ovary. Such exchange is essential...
